小鼠的腎臟被切除5/6后,全身的血液濾過功能將由殘余的腎單位承擔(dān)。劇烈的血流動力學(xué)改變會直接導(dǎo)致腎小球高濾過,進(jìn)而導(dǎo)致腎小球硬化和殘余腎單位的破壞,出現(xiàn)以腎小球肥大、硬化等為主要特點(diǎn)的慢性腎衰竭的腎間質(zhì)纖維化病變。
為保證小鼠存活率,常采取兩步法對小鼠腎臟進(jìn)行切除:
1、適應(yīng)性喂養(yǎng)1周后,麻醉小鼠,并切除小鼠左腎2/3;
2、一期手術(shù)7天后,切除整個(gè)右腎。
